Reduction of phenylpyruvic acids to phenyllactic acids in mammalian tissues.

چکیده
Evidence is presented that aromatic ol-keto acid reductase accounts for at least 80% of the total activity responsible for the reduction of aromatic a-keto acids to their corresponding phenyllactic acids in mammalian tissues. The remaining activity can be attributed to lactic dehydrogenase, which is in keeping with the known broad substrate specificity of this enzyme. The relative contribution of aromatic a-keto acid reductase and lactic dehydrogenase in a variety of mammalian tissues was determined with the use of sodium dodecyl sulfate, a potent inhibitor of lactic dehydrogenase.

The hydroxylation of phenylalanine and antipyrine in phenylpyruvic oligophrenia.
In normal individuals the major portion of ingested phenylalanine is converted into tyrosine. In the disease phenylpyruvic oligophrenia, however, ingested phenylalanine is converted mainly into phenylpyruvic and phenyllactic acids (l), which are excreted in the urine. This phenomenon has been ascribed to the inability of the body to convert phenylalanine to tyrosine (2) and a consequent shuntin...

An NAD-dependent D-lactate dehydrogenase (D-nLDH) of Lactobacillus bulgaricus ATCC 11842 was rationally re-designed for asymmetric reduction of a homologous series of α-keto carboxylic acids such as phenylpyruvic acid (PPA), α-ketobutyric acid, α-ketovaleric acid, β-hydroxypyruvate. Compared with wild-type D-nLDH, the Y52L mutant D-nLDH showed elevated activities toward unnatural substrates esp...

The mechanism of the intermediary metabolism of the more complex amino-acids still remains obscure. It is generally CODceded that, as far as concerns the a-amino group, deamination is accompanied by oxidation with the formation of cu-keto or cr-hydroxy derivatives of the amino-acids. While the manner of oxidation of the cyclic portions of such amino-acids as contain the benzene or other rings i...
متن کاملEnantioselective Biosynthesis of l-Phenyllactic Acid by Whole Cells of Recombinant Escherichia coli.
BACKGROUND l-Phenyllactic acid (l-PLA)-a valuable building block in the pharmaceutical and chemical industry-has recently emerged as an important monomer in the composition of the novel degradable biocompatible material of polyphenyllactic acid. However, both normally chemically synthesized and naturally occurring phenyllactic acid are racemic, and the product yields of reported l-PLA synthesis...
